Mycoplasma Testing Service (Conventional PCR)
Mycoplasma contamination is most widespread contamination found in cell culture. The small size of Mycoplasma makes them invisible to the naked eye as well as conventional light microscope. Further, presence of Mycoplasma in cultures may not produce visible signs of contamination but can have adverse effects on cell line growth and characteristics. Subsequently, Mycoplasma contamination may influence the experimental results and its reproducibility. In order to achieve the reproducible results from cultured cells, it is advised to test cells for Mycoplasma contamination.
PCR based detection method is widely used for reliable results by reputed organisations and international cell banks. It is one of the most rapid, sensitive and convenient method. The primer set is specific to the highly conserved 16S rRNA coding region in the Mycoplasma genome. The detection spectrum includes most of the species of Mycoplasma usually encountered as contaminants in cell cultures.
Special Note for DNA Sample Preparation:
The DNA sample from cell line to be tested for Mycoplasma contamination should be derived from:
1. Cultures grown without antibiotics for several passages (since it affects the test sensitivity).
2. Continuous cultures (at least one or two passages after thawing) and 80-90 % confluence at the time of DNA extraction.
