Research

Role of inflammation in autoimmunity and tolerance

Inflammation is the complex set of reactions involves a set of cytokines, chemokines, adhesion molecules and various cell types. Inflammation can happen in any organs or tissues in response to traumatic reaction, infection, early cancer, post-ischemic, toxic or autoimmune injuries. This helps in the generation of pro-tumorogenic environment, promoting angiogenesis and lymphangiogenesis, stimulating DNA damage, remodeling of extra-cellular matrix protein (ECM) to facilitate metastasis and evading the host defense mechanism. However, the cellular and molecular mechanisms of inflammation that control the differentiation and function of subsets of immune cells (T cells, B cells and DCs) in the inflamed microenvironment is not well studied.
We are investigating how various cytokines, chemokines and cell adhesion molecules present in inflamed microenvironment induces genome-wide epigenetic modifications and gene expression in different subsets of CD4 T cells (Th1, Th2, Th17 and Tregs). Using skin transplantation and autoimmune disease (Experimental Colitis and Experimental Autoimmune Encephalitis, EAE) animal models, we are investigating how immune cell differentiation and functions are affected in vivo in the inflamed tissues and secondary lymphoid organs.
